By executing EWZ5 t-code in SAP, all the user can be locked at the same time in SAP.

By executing su10 t-code in SAP,and click on the authorization tab and select the users (which users you want to locked) next click on the lock option .by doing this process all the user can be locked at the same time in SAP.
