hplc?

Answer / keerthi achanta
for method development, better to use gradient method
because we are try to develop the method by changing the
different composition of mobile phase.
once if we know the method then we can use isocretic method

I think
Isocrtic same composition mobile phase but some impurites not eluted
Gradient diffrent mobilephases(acn:buffer) impurites eluted good

Sort n simple, large no.of impurity separated in gradient but it is take laser analysis time, while in isocratic also large no.of impurity got separated but it takes too much time kp to 2 to 5 our depends compound / impurities nature.

Elution strengths of different components towards mobilephase is different.Hence, elution purpose different type of mobilephases used as gradient system

Pick Merging problem of chromatography thats why use gradient system.
Easily elute the pick and no interference in Chromatography then use isocratic.

A separation in which the mobile phase composition remains constant throughout the procedure is termed isocratic (meaning constant composition). the mobile phase composition does't have to remaian constant. A separation in which the mobile phase composition is changed during the separation process is described as a gradient elution. Then isocratic method used fro constant composition position of mobile phase and gradent method used for mobile phase is low to high concentration.
