autoclave is the machine which is used for sterilization of
glassware, small equipment and media i.e kill micro-organism
and their spores at 121 Celsius temp and under pressure of
100kpa (15 lbs).
Principle: The basic principle of autoclave is the
sterilization of material by using steam and pressure
because steam have greater penetration power so it can
easily penetrate in solid material so it is the best way to
sterilize the material.

sterilizatio in autoclave is complete only when each and every part of the autoclave reaches 121 degree centigrade.this happens when air in the autoclave is completely replaced by the steam.Air is completely let outside when temperature in autoclave reaches 100 degree centigrade by opening vent for one minute.

the main principle of steam sterilizer(autoclave) is irreversible denaturation of enzyme and protein structure under high vacuum and high pressure.(when steam is saturated ,it has maximum capacity for condensing out,when saturated steam condense it gives the latent heat to the material on which it condense. the heat up of objects being sterilized at temperature of 115 to 135 degree Celsius,
the condensed water also assists in the sterilization process by hydrating the microorganisms present and allowing them to be easily killed.

As the pressure increases so would the temperature. So at 15psi the temperature would be 121C which is higher than the boiling point of water and would destroy most microorganisms and their spores.

Autoclave works on Gay Lusaac's Law. If you solve the equation it says Pressure is directly proportional to temperature unlike Boyle's law where its inversely proportional. So as we increase the saturated steam pressure inside the chamber to get 121 degrees Celsius which ultimately increases the temperature of liquid to be sterilized inside the chamber without converting it into steam. (Please refer water phase diagram for better clarity) so due to water phase activity liquid media can attain 121.1 degrees without turning into steam since boiling point of water is 100 degrees. So, as per phase diagram of water if you take the temperature of autoclave chamber to 121.1 degrees by maintaining the chamber pressure at 1.1 bar or 15 PSI, it will serve the purpose of sterilization (Without making the liquid turn into steam). I hope this gives you idea.
