What is the principle of Autoclave

Answer / navneet
The main principal of autoclave is when water boiled/heated in a closed container, saturated steam is produced under pressure. According to Boyle's law ,when volume of the steam is kept constant, the temperature is directly proportional to pressure. At 15 lbs pressure 121°C temperature can be obtained. Moist heat coagulates cell protein of the microorganisms and thus kills all the living entities (including spores) in 1in 15-20 min. Hot saturated steam is efficient means of destroying microorganisms, since it penetrate rapidly and gives up its latent heat as it condenses on the cooler object.

Answer / ami
autoclave is as instrument which uses moist heat generated at 121 degree centigrade for 20mins at 15lbs pressure to kill all microorganisms. Moist heat is preferred over dry heat as it has higher penetration power.

Answer / monika yadav
due to increase in pressure of the steam formed in object
surrounding the boiling point of water get increased (
according to gas law PV=NRT ) now at high pressure more
energy is required to escape the water molecules from water
surface .
scientists had find out that 15 psi pressure will raise the
boiling point to 121 c which is sufficient to even kill the
endospores of microbes i.e . making the object sterile

The principle of the autoclave of steamed sterilizer is that water boils when its vapour pressure equals that of the surroundings atmosphere.Hence, when pressure inside a closed vessel increases, the temperature at which water boils also increases.

An autoclave is piece of equipment that is usually used to
sterilize objects and supplies by subjecting them to high
pressure saturated steam at 121 °C or more, typically for
15 to 20 minutes depending on the size of the load and the
contents. The sterilization occurs by three mechanisms:
1. temperature
2. pressure and
3. thermal oxidation.
In this temperature all the viruses kill and protein also
denature completely.

Answer / sangya agarwal
it is basically wet heat sterilisation, used to steam
sterilise equipments at 121 c and 15lbps for 15 to 20
minutes, is more effective than the use of disinfectants
and is also an alternative to incineration.

Answer / satya prakash singh
The principle of autoclave - steam above 100°C or saturated steam has a better killing power than dry heat. Bacteria are more suseptible to moist heat as bacterial protein coagulates rapidly. Saturated steam can penetrate porous material easily. When steam comes into contact with a cooler surface it condenses to water and liberates heat to that surface. The condensed water produces moist conditions for killing the microbes present.

Answer / neha sahu
autoclave provied heat in the form of saturated steam under
pressure. It gives rapid heating, Penetration and moisture
in abundanance which causes coagulation of protein.
