Colcemid is a drug that inhibits microtubule formation by binding to tubulin, the protein subunit of microtubules. It is commonly used in cell culture to arrest cells in mitosis, making it easier to isolate chromosomes for analysis. In primary cultures, Colcemid is added to the culture medium for a few hours before harvesting the cells to collect metaphase-arrested cells that are rich in chromosomes. The collected cells can then be processed for chromosome spreads and staining with dyes such as Giemsa or fluorescent dyes.
