The United States Pharmacopeia (USP) provides two methods for tapped and untapped density to account for the variation in particle size distribution, compression force, and other factors during tableting. The tapped density method (USP <257> Appendix <1>) is used when particles are highly compressible, such as microcrystalline cellulose or lactose, while the untapped density method (USP <908>) is typically applied for less compressible materials like sugar and starch. The tapped density method involves tapping the powdered substance in a cylindrical mold under standardized conditions to simulate compaction during tableting, whereas the untapped density is determined directly from loose powder without any compaction.
