Why we are maintaining the autoclave temperature at
121degree centigrade and pressure at 15lbs? Why we are not
maintaining the temperature and pressure above or below
121degree centigrade and 15lbs? higher temperature also
kills microbes. then why we are not maintaining it above
121degree? what is the reason?

bcoz the approx 120 degree is the highest temp which the spore
of thermophilic bacteria (Bacillus sterothermophilus)can
withstand.
Hence for autoclaving temp is kept 121 so as to kill even the
spores is killed.
Now why the temp is not increased from 121 is bcoz at higher
temp the media, or other nutritious material which u r
autoclaving can get denatured.

S.thermophillus bateria is heat stable bacteria, this one
also killed at 121 degree c and 15 lb p by coagulate the
protein of bacteria. thats by we maintaine the temp 121 or
15lb pressrue. At higher temperature nutrient
denatured.

we are maintaining auto clave temperature at 121degree centigrade at 15lbs pressure this is for complete destroy spores.If we keep above 121degree centigrade we have to decrease the time that is 125degree centigrade for 10minutes and 130degree centigrade for 3minutes. If we keep above 130degree centigrade there the nutrients will denatured.
