Why we use caffeine for HPLC calibration in Detector wave ength accuracy test..

Answer / anil ramnani

Caffeine is used because it gives maxima at 272 and 205 nm and minima at 245 nm Wavelength, and it's preparation is stable at room temperature, and it is soluble in water and easily available.

Why we use caffeine for HPLC calibration in Detector wave ength accuracy test..

Answer / kishanpal singh, fare labs pvt

1. caffeine easily available in market.
2. In general we all use caffeine as a standard for
calibration of HPLC specially for (Calibration of Wave
Length Accuracy) since it will scan the whole of the UV
range i.e It gives a multiwavelength response like
first maxima at 205 nm
second maxima at 273 nm &
minima at 245nm
3. It is very stable, we can use a long time for analysis
i.e. it is not degradable
4. In this case we can use Erbium Perchlorate which will
scan the whole range(200 nm to 600 nm).It is stable and
little costly but not easily available in market.

Why we use caffeine for HPLC calibration in Detector wave ength accuracy test..

Answer / k.rajasekhara reddy

It is different wavelengths two maxima and one minima wavelengths 204, 272,245nm. this is no degradation due to environmental factors easily available and low cost in market

Answer / suryabhan datir

caffeiene having different wavelents it having 2 maximas
and 1 minima.
1st maxima is 205nm
2nd maxima is 273nm
minima is 245nm
and it is primary reference standard and also suggested in
pharmacopiea. It is easily soluble in water. It shows
complete absorbance at multiwavelength

My question about gas chromatography sulfur chemiluminsecence detector. I test unknown sample gas by GC-SCD (calibrated ) and the result of *H2S is 279 PPM , *but when I test the same sample with the GC-TCD (calibrated ) the value of *H2S is 2500 PPM . I'd like to inform you that both GCs are calibrated and have very good operation conditions with stable parameters . the question is if the sample gas with higher H2S over detection limits of SCD detector (1000 ppm). why the result it 279 ppm Best regards
