Cell counting using absorbance or turbidity is a common method to determine the concentration (cell density) of microbial cultures. The procedure involves measuring the optical density (OD) at a specific wavelength (usually 600 nm for bacteria).n
1. Sample preparation: Prepare a dilution series of the cell suspension and transfer an aliquot to a sterile cuvette.n
2. Measurement: Read the OD using a spectrophotometer and record the value.n
3. Calibration: Calibrate the OD against a standard curve prepared with known cell concentrations (usually prepared from a cell suspension of known concentration).n
4. Analysis: Plot the OD values on the y-axis against the logarithm of the cell concentrations on the x-axis to obtain the standard curve. Determine the cell density of the unknown sample by interpolating the OD value from the standard curve.
