Ingredients are
KH2PO4 0.5 g
MgSO4.7H2O 0.5 g
Citric acid 2.0 g
Ferric ammonium citrate 0.05 g
Glycerol 60 ml
Asparagine 4.0 g
Distilled water to 1000 ml.
Dissolve the reagents, except for the glycerol, in 300 ml
water using heat.
Once dissolved, add the glycerol and make up to 1 liter
with distilled water. Check pH and, if necessary, adjust to
pH7.4 with 1 M NaOH.
Autoclave at 121 oC for 15 min.
It has been reported that the addition of ZnSO4 (0.1 ml of
1% solution) and Triton WR1339 (5 ml of 5% solution)
improves growth of Mycobacterium. tuberculosis .

Dissolve asparagin separatly in 100 ml distilled water
using magnet stirer & heat upto 56 Deg C
After dissolve the other reagents, except for the glycerol,
in 300 ml water one by one,using magnet stirer , add the
asparagin solution and after 5 minuts add glycerol and then
make up to 1 liter.
