No,Tester cannot write Test Cases by refering Design Doc.
Tester can write Testcases by ref. FRS,SRS etc but nt by
refering Design doc.

There is no need of design documents for a tester to write
the test cases. They would only require SRS document and
domain knowledge which would be shared with them by the
domain experts.
