To culture bone marrow mononuclear cells (BMMC), obtain bone marrow aspirate from a donor, layer it onto Ficoll-Paque Plus, and centrifuge at appropriate speed. The mononuclear cells will be found at the interface between the bone marrow and the Ficoll. Wash the cells, resuspend them in culture medium (such as Iscove's Modified Dulbecco's Medium with 10% fetal bovine serum), and plate them for further culturing.
