C2C12 cells, a mouse myoblast cell line, are commonly used to study skeletal muscle development and differentiation. To induce differentiation, C2C12 cells are typically cultured in high serum (usually 10-20% fetal bovine serum) conditions for several days until they reach confluence. At this point, the cells are switched to low serum (typically 2%) or serum-free media containing various growth factors such as insulin and dexamethasone, which promote muscle differentiation. Over a period of several days, C2C12 cells will begin to fuse together and express muscle-specific markers like myosin heavy chain (MHC), indicating successful differentiation.
