Bacterial two-component response regulators are essential components of bacterial signal transduction systems. These regulatory proteins function in a two-step process, where the initial sensing of environmental signals is performed by sensor kinases and the subsequent transcriptional regulation is carried out by the regulator proteins. The regulator protein can exist in two conformations: an auto-phosphorylated active conformation and an un-phosphorylated inactive conformation. Upon sensing a specific environmental signal, the sensor kinase undergoes autophosphorylation and then phosphorylates the response regulator. This phosphorylation event leads to a conformational change in the response regulator that allows it to interact with DNA and modulate gene expression.
