Separate the nucleoid (portion containing nucleic acids) of
the prokaryote by cell lysis and differential
centrifugation. After that, treat the extract with DNAse to
remove any trace of DNA along with the RNA. The RNA thus
obtained is mRNA.

first we have to isolate complet RNA THEN apply that on
oligo dT Cellulose column then you can get your m-RNA

gradient then separate out Genetic material and then use
DNAase enzyme to lyse DNA then extract rna by centrifugr or
gel electrophoresis method

prokaryotes do not produce mRNA... this is produced in the
riobosomes of Eukaryotes... prokaryotes produce rRNA which
can be separated by lysing the Shine-Dalgarno sequence with
BBNT and then applying a
dideoxyoligonucleotidephosphohydroxylase to the upstream
portion of your okazaki fragments
