How will you select mobile phase in HPLC Method Development..

Answer / maulik

based on the pka value of compound. if ur compound have pka
is about 3. u can select ur mobile phase about ph 5-14.but
due to the less column selectivity in basic media u can
select 5-8 pH. preferred by the different salt buffering
capacity.

How will you select mobile phase in HPLC Method Development..

Answer / rishabh

First of all check your compound solubility in which solvents then after that based on solubility the Mphase selection becomes easy if compound is soluble in aqueous medium check the pH of that medium than after further procced if not then check the solubility in organic solvents on basis of solubility and pH of the Mobile phase. Both Mobile phase selection and stationary phase are selected.
