TIE is the answer, for if you would tell that your word is DUE it cannot be easily determined for there are words that sounds like due e.g., dew, but they have different no. of vowels while in TIE, there are 2 vowels and also the words that sound like it, e.g., THAI, has also 2 vowels

The question isn't written correctly. It should be -
"if I were to tell you any one 'LETTER' of secret word, then
you would be able to tell me the 'number of vowels' in my
secret word."
In that case answer would be - DUE,
because in the above set of letters(A,E,I,O,U,M,T,D) every
letter except 'U' has been used twice in two different words
and hence if given would give two possible choices.
