If the pump is filled with air instead of water, the air is too elastic, (too compressible) for the typically designed pump to work. An exception would be a self priming pump (which is more efficient at pumping air). However, Even a perfectly designed self priming pump can not draw water from more than 32 feet because of the limitation of atmospheric pressure.

water pump requires the inlet pipe to be filled with water
for it to function . Due to various reasons the tube could
do dry (like running the pump dry). By pouring the water, we
are basically filling this tube. Once the tube is full, its
ready to pump water again. This activity is called priming a
pump
