There are 3 conformation observed on 1% agarose gel run of isolated Plasmid DNA, they are -- supercoiled circular DNA, nicked supercoiled circular DNA, and relaxed circular Dna. If the isolated plasmid DNA is not treated with Rnase enzyme then another band is observed in agarose gel known to be DNA masked with RNA. Read lehninger and stryer.
