What should be the calibration procedure for wavelength
accuracy in case of HPLC?
Answer Posted / ankama rao.puppala
wavelength accuracy was done by the following reagents
1.Caffeine
2.Erbium Perchlorate
3.benzophenone
In case of Caffeine
we check the absorbance(Area)the following nanometers
200nm-210nm(205nm is the maximum absorbance)
270nm-280nm(273nm is the maximum absorbance)
240nm-250nm(245nm is the miminum absorbance)
we got the above parameters wavelength accuracy was
passed...
In the above Caffeine was best reagent becos of
it has Two maxima(205nm, 273nm) and one minima(245nm)
More stable, Cheaf cost more over it is recomanded by
Pharmacopeia.
