The most convincing test is based on fine structure of cell
wall visualised by thin-sectioning e-micrscopy (outer and
cytoplasmic membranes, separated by thin layer in G-, and
cytoplasmic membrane covered by thicker peptidoglican layer
in G+). Gram-variability by afore tests is known for some
bacteria
