The basic principles involve coating a solid surface with the antigen, adding a solution containing the substance of interest, washing away unbound substances, introducing an enzyme-labeled antibody specific to the substance, and finally adding a substrate that reacts with the enzyme to produce a measurable signal. The intensity of the signal corresponds to the amount of the substance in the original sample.
