How to calculate protein percentage in lowry method?
Answer Posted / Alka Chauhan
The Lowry method is used to measure the total amount of protein in a sample. To calculate protein percentage, follow these steps:
1. Measure the absorbance of your protein sample at 570 nm using a spectrophotometer.
2. Prepare a standard curve by measuring the absorbance of known concentrations of BSA (bovine serum albumin) and plotting them against their concentrations.
3. Determine the concentration of your protein sample from the standard curve.
4. Weigh your protein sample to find its mass (m1).
5. Weigh a known weight of another substance (like water) that is not included in the Lowry method but has the same volume as your protein solution (m2).
6. Calculate the total mass of the sample and the other substance (m_total = m1 + m2).
7. Calculate the protein percentage using the formula: Protein Percentage = (mass of protein / m_total) * 100
