Isolation of the outer membrane of a cell involves several steps, including mechanical and enzymatic lysis to break open the cells. The resulting crude membrane fraction is then separated from other organelles by differential centrifugation. The pellet containing the outer membrane can be further purified using techniques such as detergent solubilization, ultracentrifugation, or chromatography.
