The extraction of red blood cells (RBCs) from whole blood is typically performed using anticoagulants to prevent clotting. The process involves centrifugation of the blood sample at low speeds, resulting in a buffy coat containing white blood cells and platelets at the bottom, RBCs in the middle, and plasma at the top. The RBC-rich layer is carefully removed and washed multiple times with isotonic saline solution to remove residual plasma. After washing, the RBC pellet can be resuspended in an appropriate buffer for further analysis or storage.
