Double Thymidine Block (DTB) is a synchronized cell culture technique used to arrest the DNA synthesis phase (S-phase) in Hela cells (a human cervical cancer cell line). The process involves exposing the cells to thymidine, a nucleoside analog that inhibits DNA synthesis. After the first thymidine treatment, the cells are washed and allowed to continue growing before being re-exposed to thymidine for a second block. This technique is useful for studying cell cycle kinetics and the regulation of DNA replication.
