1. Inoculation: A loopful of culture from an overnight growth is inoculated into a fresh medium containing glucose and minerals.
2. Incubation: The inoculated culture is incubated at optimal temperature (usually 28°C) for a specific period (typically 48-72 hours).
3. Extraction: After the incubation period, the cultures are extracted with an organic solvent (such as chloroform or ethyl acetate) to isolate the rhamnolipids.
4. Measurement: The extracted rhamnolipids can then be measured using various methods such as spectrophotometry or thin layer chromatography (TLC).
