Heat the DNA upto 94 degree celsius. If it is absorb more UV light at 260 nm, that will indicate that DNA is double stranded because of base stacking DNA can absorb more light (called as hyperchromic effect). If the absorabance will same at 260 after heating, then DNA will be single stranded.
