what buffer you can select for acidic nature analyte and
how ?
Answer Posted / shyam kumar
For an acidic nature compound u should use only acid pH
buffers only why because acid compounds are showing more
hydrofobicity in acid pH, if the sample is having more
hydrofobicity it retains on column and it gives good peak
shape and advisable retention time,
if we do acid samples in basic pH buffers the sample
undergoes ionisation meanwhile increases the hydrophilic
nature dueto this hydrophilic nature the sample not retains
on the column. u must select a acid pH to analyse the acid
samples.
