northern blotting is simply an extension of southern
blotting. In northern blotting RNA is transferred from gel
to membrane instead of DNA.
The RNA sample are most commonly separated on agarose gel
containing formaldehyde as a denaturating agents.RNA is
transfer to diazobenzyloxymethyl(DBM) paper or nylon
membrane through capillary action or vacuum blotting
system.RNA blot is hybridized to either RNA or DNA labelled
probes and detected by x-ray film.
This technique is extremely useful in studies of gene
expression They can be used to determine whether a
particular gene istranscribed of an organism or only in
certain tissue.
They can also be used to study the temporal pattern of
expression of individual genes during growth and
development.
