Potency is a measure of drug activity expressed in terms of
the amount required to produce an effect of given intensity.

Purity is a measure of the amount of API present in a sample
compared to those of related substances, impurities,
residual solvents, etc.

we are calculating by area normalisation.

while Potency is content of that substances in the sample
and calculating it on dried basis ie. LOD, water content ,
residual solvent.

residual solvents, etc. now another term is purity is carried out by HPLC chromatographic purity and is % area of Main Peak where all peak integrated including all
known peaks due to product

i am not in favored of potency calculated on basis of assay
rather than purity because assay calculated against another
std and there will be a possibility of no. of errors in
define correct assay.
