first find out the solubility of the drug and then pka of the drug and absorbtion maxima of the drug on uv.And then start the trails by using different ratio of aq and organic phase.If we dont get sufficient parameters then go foe the buffer and it should have a ph of +/- 1 of pka of the drug.Then go for the trails.
