To define new regions in Drupal, follow these steps:
1. Go to Appearance > Themes and select your active theme.
2. Click on the 'Configure' link under the theme name.
3. Navigate to the Regions tab.
4. Add a new region by entering a Region Machine Name and a Region Label.
5. Save the changes.
