Can nybody tel me the how much a Manual tester should know
about Database and its connections with the web application?

Knowledge on database does not depend on web/windows/...
application.For a tester, on a scale of 10 you should at
least score 5.You should be familiar with DDL,DML
statements,JOINS and a basic knowledge on SP's.
