The Riboflavin (vitamin B2) assay is a simple microbiological method used to detect and enumerate Enterobacteriaceae. The test involves inoculating a sample onto a selective medium containing lactose, bile salts, and fuchsin dye. In the presence of air and light, riboflavin produced by Enterobacteriaceae decolorizes the red fuchsin dye, turning the medium yellow or greenish-white. The intensity of the color change correlates with the number of Enterobacteriaceae present in the sample.
