If we are giving the missing authorization(T-
codes/Authorization objects) to the User. User no need to
log off from the System.It will work without logging off
from the system.

Yes, User must have to loggoff and relogin to activate
his/her newly acquired TCodes or authorization object.

If we are assigning missing authorizations(T-codes/Authorization Object) there is no need of user to log off and login again.

While changing authorization data under authorization tab in PFCG(T-Code),we generate and reconcile the profile of that particular role which takes effect immediately on all the users which are added to this role.
